帕金森模型大鼠海马区Pitx3和Nurr1基因表达情况的实验研究

摘要 目的制作帕金森(PD)大鼠模型,研究其海马区Pitx3和Nurr1基因表达情况。方法纹状体内注射6-羟多巴胺制作大鼠帕金森模型,于不同时间点处死,免疫荧光方法检测Pitx3、Nurr1基因在帕金森大鼠海马区的表达。结果同正常组、假手术对照组相比较,PD大鼠海马区的Pitx3+细胞,Nurrl+细胞和Pitx+/Nurrl+细胞数在模型成功后的第3、7天明显增加,第14天后逐渐减少,第28天后恢复正常水平。结论Pitx3、Nurr1基因在帕金森模型大鼠的海马区大量表达,可能与帕金森大鼠新生的神经干细胞的增殖有关。 Objective To investigate the gene expression of Pitx3 and Nurrl in hippocampus of hemi-parkinsonian rats. Methods Rat model of Parkinsons disease was made by injecting the 6-hydroxydopaminin in the striatum.The rats were killed at different time points,and their brain were taken out. Using the methods of immunofluorescence,the gene expression of Pitx3 and Nurrl in hippocampus of hemi-parkinsonian rats were detected. Rsults Campared with the controls,the number of Pitx3-positive cells,Nurrl-positive cells,and Pitx3-Nurrl-positive cells increased strikingly at 3 and 7days in the hippocampus,then declined at 14 days and almost reached the normal 28 days after. Conclusion Pitx3 and Nurrl genes were strongly expressed in hippocampus of hemi-parkinsonian rats, which might be related to the proliferation of neural stem cells.
