摘要
目的:考察四红祛斑汤对肝郁脾虚型黄褐斑的预防作用及其机制,为临床治疗提供指导和依据。方法:在以黄体酮注射+慢性束缚+紫外光照射建立肝郁脾虚型黄褐斑小鼠模型的同时,给予四红祛斑汤灌胃预防。通过比较正常组、模型组和预防组HMB45标记的小鼠皮肤黑色素细胞内黑色素颗粒的变化,评价该复方的预防作用。并通过观察小鼠皮肤一氧化氮合酶(NOS)和酪氨酸酶mRNA的表达变化,探讨该方预防黄褐斑生成的机制。结果:四红祛斑汤预防组的小鼠皮肤黑色素细胞内黑色素颗粒较模型组明显减少,对NOS和酪氨酸酶mRNA的表达也有显著抑制。结论:四红祛斑汤对于肝郁脾虚型黄褐斑小鼠模型具有较好的预防作用,其作用机制可能是该方可以预防皮肤黑色素细胞的NOS和酪氨酸酶mRNA的表达,以减少皮肤黑色素的生成。
Objective: To observe the prophylaxis effect and mechanism of Sihongqubantang on the chloasma model of stagnation of liver-QI with deficiency of the spleen syndrome, and provides directions and basis for clinical treatment. Methods: When building the mouse models of stagnation of liver-QI with deficiency of the spleen syndrome by injecting Progesterone and chronically constrainting and exposure to UV rays, the mice were intragastrically administrated with Sihongqubantang to prevent the formation of chloasma. Then the prophylaxis effect of our complex prescription was appraised by comparing the changes of melanin granules in the melanocytes labeled with HMB45 in normal group, model group and prevention group. And then the mechanism of prophylaxis effect of our complex prescription on the formation of chloasma was explored by the changes of levels of nitricoxide synthase (NOD) and tyrosinase mRNA in the skin of mice. Results: The number of melanin granule in melanocytes of the mice in prevention group was significantly decreased than that in model group, and a considerable inhibitory effect was also observed on the expression of both NOS and tyrosinase mRNA. Conclusion: Sihongqubantang shows a prophylaxis effect on the chloasma mouse model of stagnation of liver-QI with deficiency of the spleen syndrome, and its possible mechanism is: our complex prescription can prevent the expression of nitricoxide synthase (NOD) and tyrosinase mRNA in melanocytes and further decreases the production of melanin in skin.
