高效液相色谱法测定小鼠血浆中芦荟大黄素的浓度

摘要 目的:建立以高效液相色谱法测定小鼠血浆中芦荟大黄素浓度的分析方法。方法:取0.3ml小鼠血浆,二氯甲烷萃取;HPLC采用Agilent 1100高效液相色谱仪系统,以Symmetry Shield RPC18为色谱柱,流动相为甲醇-水-醋酸(65∶35∶0.2),并用荧光法测定(λex=410nm和λem=510nm)。结果:芦荟大黄素的保留时间为11.7min;芦荟大黄素浓度在10~1000ng/ml(r=0.999)的范围内具有良好的线性关系,最低检测限(LOD)和最低定量限(LOQ)分别为4.5ng/ml和5ng/ml。结论:本方法简便,准确,灵敏,特异性强,重现性好,可用于血浆中芦荟大黄素的测定及药动学研究。 Objective: To develop a HPLC method for the determination of aloe emodin (AE) in mouse plasma. Methods: 0.3 ml of plasma was taken and extracted by dichloromethane. The Agilent 1100 HPLC system was used with the Symmetry Shield RP C18, a mobile phase was methanol-water-acetic acid (65:35:0.2) and fluorescence detection was set at λex=410 nm and λem=510 nm. Results: The retention time of AE was 11.7 min, There was a good linear relationship at the range of 10 -1 000 ng/ml for aloe emodin (r=0.999), the LOD and LOQ were 4.5 ng/ml and 5 ng/ml, respectively. Conclusion: The method is simple, accurate, sensitive, with high specificity and good reproducibility, it can be used for the determination of aloe emodin and pharmacokinetics study of aloe emodin in mice.
