利用桑叶和麸皮固体发酵毛云芝菌产漆酶的培养基优化研究

摘要 采用响应面法对毛云芝菌固体发酵产漆酶的培养基进行优化。首先运用部分因子筛选实验(FFD)筛选出对发酵培养基影响较大的重要因素,分别为桑叶与麸皮比例、草酸铵、葡萄糖;然后用最陡爬坡实验逼近最佳响应面区域,最后通过Box-Behnken实验设计,利用SAS进行回归分析和岭脊分析,得到主要因素的最佳比例及浓度。在优化培养基的条件下,固体发酵培养酶活较初始培养基提高了10倍。 Response surface methodology(RSM) was employed to optimize culture medium for the laccase production in solid cultivation by Coriolus hirsutus. A fractional factorial design (FFD) was used to screen out the important components for the fermentation substrate. It is inverse proportion of mulberry leaves and bran, oxalic acid ammonium and glucose. The path of steepest ascent was used to approach the optimal response re- gion. The proportion and concentrations of the significant factors were optimized consequently through Box -Behnken experiment design and using regress and ridge analysis. As the result, the production of laccase under solid culture with optimal medium was 10 times higher than under the original medium used previously.
