依曲替酸和黄芪对HaCaT细胞增殖及细胞周期的影响

摘要 目的:研究依曲替酸和黄芪对HaCaT细胞增殖及凋亡的影响。方法:采用MTT比色法检测依曲替酸和黄芪对HaCaT细胞增殖的影响;应用流式细胞仪检测细胞周期和凋亡并于透射电镜下观察凋亡细胞形态。结果:依曲替酸和黄芪均对HaCaT细胞增殖有明显的抑制作用;在一定浓度范围内其抑制率呈时间和剂量依赖性,但黄芪的抑制率较依曲替酸为低。细胞明显阻滞于G1期,有明显的凋亡峰出现,电镜下见典型的凋亡细胞形态学特征。结论:依曲替酸和黄芪均可抑制HaCaT细胞的增殖并诱导其凋亡,提示二者治疗银屑病机制可能与此有关。 Objective: To investigate the effects of acitretin and Huangqi injection on proliferation, apoptosis of HaCaT keratinocytes. Methods: The effects of acitretin and Huangqi injection on the proliferation of cultured HaCaT cell were measured by MTT assay. Cell cycle and apoptosis were analyzed by flow cytometry. Morphological changes were observed under transmission electron microscope. Results: Both acitretin and Huangqi injection inhibited the proliferation of HaCaT cells in a time - and concentration - dependent manner, but the effect of Huangqi was weeker than that of acitretin. Both drugs arrested HaCaT cell in G, phase and induced prominent apoptosis. Under transmission electron microscope morphological changes of typic apoptosis were found. Conclusion: Both acitretin and Huangqi injection can inhibit the proliferation of HaCaT cells and induce apoptosis, which suggest the antipsoriatic effects of the two drugs.
