单细胞凝胶电泳技术检测激光对小鼠脾淋巴细胞DNA的损伤程度

Single-cell Electrophoresis: a Method to Detect DNA Damage Grade of Mouse's Spleen Lymphocytes Irradiated by Laser

摘要 目的:通过单细胞凝胶电泳技术检测不同强度激光照射对小鼠脾淋巴细胞DNA的损伤程度。方法:使用能量密度为600μJ/cm2的YAG激光分别照射小鼠脾淋巴细胞15、25、35个脉冲后,采用单细胞凝胶电泳法检测实验组和对照组小鼠脾淋巴细胞DNA的损伤程度,并用CASP软件进行定量分析。结果:激光照射脉冲数与淋巴细胞DNA损伤程度呈正相关。结论:单细胞凝胶电泳技术可以作为一种新的方法用于激光对生物组织DNA的损伤检测。 Objective:To detect DNA damage grade after spleen lymphocytes of mouse were irradiated by laser. Methods: Lymphocytes were irradiated by YAG laser with the energy density of 600μJ/cm^2 for 15 pulse, 25 pulse, 35 pulse respectively. Then single-cell gel electrophoresis (SCGE) was used to detect damage grade of DNA of the test group and the control; CASP software was introduced to make quantitative analysis. Results: High intensity laser irradiation led to severe damage of mouse's spleen lymphocytes DNA. Conclusion.. SCGE can be adopted as a new method to detect DNA damage grade of bio-tissue irradiated by laser.
