摘要
【目的】探讨苦参碱对人骨肉瘤细胞系MG-63体外增殖活性的影响。【方法】将不同浓度的苦参碱作用于体外培养的人骨肉瘤细胞,运用四甲基噻唑蓝(MTT)法检测不同浓度组药物对细胞增殖率的影响,流式细胞仪分析细胞周期的变化。【结果】随着苦参碱浓度的增加和作用时间的延长,人骨肉瘤细胞的体外增殖率逐渐下降,且大部分细胞被阻滞在S期,导致S期的细胞比例增大及G2/M期的细胞比例减少。【结论】苦参碱能明显抑制人骨肉瘤细胞的体外增殖活性,其机制可能是将细胞阻滞在S期。
[Ohjective] To investigate the effect of matrine on exosomatic proliferative activity of osteosarcomatous cell line MG-63. [Methods]Matrine in different doses were used to treat osteosarcomatous cell line MG-63 for 0,12,24 and 18h, respectively. MTT assay was employed to evaluate the influence of matrine on exosomatic proliferative activity, and flow cytometry was used to detect the changes of cell cycle induced by matrine. [Results]With the increase of the concentration of matrine and the prolongation of administration time, the proliferative rate decreased correspondingly, and many cells were blocked in the period of S, and the quantities of cells increased in the period of S and decreased in the period of G2/M. [Conclusion]The inhihitory effect of matrine on prolifcralive activity in human osteosarcomatous cell line MG-63 in vitro is significant, and its mechanism may be that most cells are blocked in the period of S.
