PSCA在胰腺癌组织中的表达及临床意义

摘要 【目的】探讨前列腺干细胞抗原(PSCA)在胰腺癌组织、癌旁组织和胰腺良性病变(慢性胰腺炎和胰腺假性囊肿)中的表达及其与临床病理特征的关系。【方法】收集10例胰腺癌患者的癌组织、癌旁组织,采用RT-PCR方法检测其PSCAmRNA表达情况;另收集34例胰腺癌及12例慢性胰腺炎和12例胰腺假性囊肿资料完整的术后患者,采用免疫组织化学法检测PSCA蛋白的表达,并分析PSCA与临床病理特征的关系。【结果】RT-PCR结果显示胰腺癌组织中PSCAmRNA表达水平明显高于癌旁组织。免疫组化分析表明,PSCA蛋白在胰腺癌中阳性表达率为59%。其表达与胰腺癌TNM分期和淋巴结转移密切相关(P<0.05),而与患者的性别、年龄、肿瘤大小等无关(P>0.05)。【结论】PSCA在胰腺癌组织表达与胰腺癌的生长和浸润转移关系密切,可以作为判断胰腺癌生物学行为的重要指标。 [Objective]To investigate the expression of PSCA(prostate stem cell antigen) in pancreatic cancer, pericancerous tissues and pancreatic innocence disease(chronic pancreatitis and pancreatic pseudocyst) and its clinicopathologic significance. [Methods]The expression of PSCA mRNA was detected by reverse tran- scriptase polymerase chain reaction (RT-PCR) in cancer samples and pericancerous tissues from 10 pancreatic cancer patients. The expression of PSCA in 34 pancreatic cancer, 12 pancreatic pseudocyst and 12 chronic pancreatitis were studied by immunohistochemistry. All cases had complete pathological data. Statistical analysis was performed to investigate the association between the clinicopathologic features and PSCA expression. [Results] RT-PCR demonstrated that the levels of PSCA mRNA in pancreatic cancer samples were significantly higher than those in pericancerous tissues. Immunohistochemical staining showed that PSCA was highly expressed in 59% of pancreatic cancer. Expression of PSCA was highly correlated with TNM stage and lymph node metastasis ( P 〈0.05). On the other hand, it was not correlated with gender, age, or tumor size ( P 〉0.05). [Conclusion]Overexpression of PSCA in pancreatic carcinoma can enhance invasion and metastasis. PSCA may serve as an important parameter to indicate biologic behavior of pancreatic carcinoma.
