摘要
目的探讨2-甲氧基雌二醇(2-methoxyestradiol,2-ME)对体外培养的SGC-7901胃癌细胞增殖、凋亡及细胞周期的影响。方法将不同浓度的2-ME不同时间作用于SGC-7901,采用倒置显微镜观察细胞的形态学变化;四甲基偶氮唑蓝(MTT)法检测2-ME对SGC-7901细胞增殖的抑制作用;流式细胞术检测细胞凋亡和细胞周期;电镜观察细胞超微结构变化。结果2-ME对SGC-7901细胞有很强的抗增殖的作用,引起大量细胞凋亡,且在一定范围内呈剂量和时间依赖性,作用组与对照组之间的差异具有统计学意义(P<0.05)。细胞周期检测发现作用组大量细胞阻滞于G2/m期G0/G1及S期细胞减少,与对照组存在显著差异(P<0.05)。结论2-ME可抑制胃癌细胞SGC-7901的增殖并诱导其凋亡。
Objective To explore the effects of 2-methoxyestradiol (2-ME) on proliferation, apoptosis and cell cycle of SGC-7901 ceils in vitro. Methods SGC-7901 cells were treated with 2-ME of different concentrations and time in vitro, morphological changes of apoptotic cells were observed under microscope; cell proliferation inhibition was detected by MTT colorimetric assay; cell cycle arrest and apoptosis rate were measured by flow eytometry (FCM). Results Exposure to 2-ME had a strong antiproliferative effect on human gastric cancer cells and caused an increase in the population of apoptotic ceils. There was significant difference between 2-ME group and control group ( P 〈 0. 05). A significant number of cells were blocked in the G2/M phases of the cell cycle,Concurrently, the population of cells in the G0/G1 and S phases decreased in SGC-7901 cell lines, detected by flow cytometry. There was significant difference between 2-ME group and control group ( P 〈 0.05 ). Conclusion 2-ME inhibits the growth of human gastric cancer cells and induces apoptosis.
