摘要
目的使用内含HCV RNA病毒样颗粒建立HCV实时荧光定量参考品,以便对实验操作进行严格的质量控制。方法使用国家一级校准品对内含HCV RNA病毒样颗粒进行含量标定,并稀释制作定量参考品。结果内含HCV RNA病毒样颗粒的RNA溶液作为HCV RNA荧光定量检测试剂盒的定量参考品,在检测方法上该参考品既可作为定量参考品,又可当作室内质控品,从样品处理开始就可以监控整个实验操作过程,对实验的可靠性提供了有效的保证。结论内含HCV RNA病毒样颗粒定量参考品的使用使临床上开展核酸扩增技术(NAT)来检测HCV病毒RNA含量定量准确,结果可靠。
Objective To accurately detect and measure the content of HCV RNA with nuclear-acid amplifying technology (NAT), fluorescence real-time quantitative standard for virus-like particles containing HCV RNA was constructed, which could control the experiment operation strictly on quality level. Methods The content of virus-like particles containing HCV RNA was measured with the national first-level calibration standard and the quantitative standard was made after diluting. Results The RNA solution of virus-like particle detection could also be used and controlled at the beginn effectively. Conclusion s containing HCV RNA as the quantitative standard for HCV RNA fluorescence as indoor quality controls. The whole experiment operation could be monitored ing of dealing with samples, which guaranteed the reliability of the experiment The use of quantitative standard for virus-like particles containing HCV RNA makes the deteetion and measurement reliable and accurate.
