摘要
目的:制备葫芦素E(cucurbitacin E,CuE)脂质体,建立HPLC测定CuE脂质体含量的方法。方法:乙醇注入法制备CuE脂质体;使用Diamonsil C18柱(200 mm×4.6 mm,5μm);流动相:乙腈-水-冰醋酸(66∶34∶0.01,v/v);流速:1 ml/min;检测波长:228 nm;柱温:35℃。结果:制得CuE脂质体的平均包封率为89.7%,在实验条件下所用辅料对CuE的测定无干扰,在3.5 mg/L^84 mg/L浓度范围内线性关系良好(r=0.9999,n=9);CuE平均回收率99.9%,RSD为0.4%(n=9)。结论:乙醇注入法可用于制备CuE脂质体;高效液相色谱法简便易行、结果准确,可用于CuE脂质体含量的测定。
Objective: To prepare cucurbitacin E liposomes by ethanol injection and to determine the content of cucurbitacin E in the liposomes by HPLC. Methods: The preparation was carried out by ethanol injection method while the determination was performed on a diamonsil C18 column (200 mm × 4.6 ram, 5 μm). The mobile phase consisted of aeetonitrile, water and acetic acid glacial (66:34:0.01 ,v/v), and the detection wavelength was 228 nm. Results: If all accessories used in the experiment did interfer the CuE determination, the average entrapment efficiency of cucurbitacin E liposomes was 89.7%. The linear range of cucurbitaein E was 3.5 mg/L - 84 mg/L ( r = 0. 9999, n = 9). The average recovery was 99.9 % and RSD was 0.4 % ( n = 9 ). Conclusion : The ethanol injection method can be used to prepare cueurbitaein E liposomes. It is a simple and accurate method to determine the contents of eucurbitacin E in the liposomes by HPLC.
