流苏石斛的离体快速繁殖 被引量:9

摘要 以流苏石斛茎段为外植体进行离体快速繁殖研究。结果显示:茎段诱导芽的适宜培养基为MS+BA2.0mg·L-1+NAA0.5mg·L-1+CH0.5g·L-1,芽的诱导率达58.3%;培养基MS+BA4.0mg·L-1+NAA0.5mg·L-1+CH0.5g·L-1可促进丛芽的形成,而丛芽增殖的适宜培养基为MS+BA2.0mg·L-1+NAA0.2mg·L-1+CH0.5g·L-1,增殖系数达6.23;试管苗生根的最佳培养基为1/2MS+IBA0.5mg·L-1+NAA0.5mg·L-1,50d后试管苗生根率可达100%。研究还发现,壮苗的适宜培养基为1/2MS+BA0.5mg·L-1+NAA0.1mg·L-1+CH0.2g·L-1,丛芽在该培养基上培养75d后,可同时完成增殖与生根程序。 Studies were carried out on rapid propagation of Dendrobiumfunbriatum in vitro, using stem as explants. The results were showed the optimal medium for inducing buds was MS+BA 2.0mg·L^-1+ NAA 0.5mg·L^-1+ CH 0.5g·L^-1, the inducing rate of buds was 58.3%; the medium MS + BA 4.0 mg·L^-1+ NAA 0.5 mg·L^-1+ CH 0.5 g·L^-1 would advanced production of clumpy buds; the MS medium supplement with BA 2.0 mg·L^-1+ NAA 0.2 mg·L^-1+ CH 0.5g·L^-1 was indicated good for propagation of clumpy buds and the propagation coefficient was 6.23; the optimal medium for root formation was 1/2 MS + IBA 0.5 mg·L^-1+ NAA 0.5 mg·L^-1, the rooting frequency was up to 100%. It was also found the optimal medium for healthy planflet was 1/2 MS + BA 0.5 mg·L^-1 + NAA 0.1 mg·L^-1+ CH 0.2 g·L^-1, which would accomplish propagation of shoots and rooting after 75 days.
