莨菪烷类生物碱与人血清白蛋白非共价结合的电喷雾质谱研究 被引量:1

摘要 利用电喷雾离子阱质谱法研究人血清白蛋白与樟柳碱、山莨菪碱、阿托品、东莨菪碱间的非共价作用,考察进样液pH值及质谱仪毛细管温度、锥孔电压等对莨菪烷类生物碱与人血清白蛋白非共价结合特性的影响.计算莨菪烷类生物碱与人血清白蛋白复合物的结合常数(K)和最大化学计量比,并根据热力学参数确定了人血清白蛋白与樟柳碱、阿托品、山莨菪碱之间的作用力类型主要为静电引力,与东莨菪碱之间的作用力类型主要为疏水作用力.电喷雾离子阱质谱法可用于莨菪烷类生物碱与人血清白蛋白非共价结合的研究. The non-covalent interaction between human serum albumin (HSA) and tropane alkaloids, such as anisodine hydrobromide (AN), anisodamini hydrobromidum (AI), atropine minimum (AT) and scopolamine hydrobromide(SC) was studied by electrospray ion trap mass spectrometry method,the influences of pH values of injection solution,capillary temperatures and cone voltages on molecular weight of the complexes were also studied. The binding constants (K) and the maximum stoichiometric ratio of the complexes are 3.11 ×10^4 mol/L/1:6 for liSA-AN,4.17×10^4mol/L/1:7 for HSA-AT,5.47×10^4 mol/L/l:5 for HSA-SC and 2.30×10^4 mol/L/1:5 for HSA-AI,respectively. According to the thermodynamic parameters of the non-covalent complexes,the main sorts of binding force are static-electricity gravitation for HSA-AN, HSA-AI and HSA-AT, hydrophobic interaction for HSA-SC,respectively. The results suggest that the electrospray ion trap mass spectrometry method is suitable for non-covalent interaction between tropane alkaloids and human serum albumin.
