Effects of rIL-2 stimulation on the activation of lymphoid subpopulations in human umbilical cord blood and adult peripheral blood

目的比较分析ｒＩＬ－２刺激对人脐带血和成人外周血淋巴细胞亚群及功能的影响。方法利用流式细胞技术对脐带血和成人外周血淋巴细胞亚群进行检测，用ｔ检验做统计学分析。结果新鲜未刺激的脐带血淋巴细胞在发育和功能方面均比成人外周血幼稚；两者对ｒＩＬ－２刺激的反应明显不同，脐带血的变化主要表现为ＣＤ４＋和ＣＤ８＋细胞亚群的成熟和记忆功能增强，即由ＣＤ４５ＲＡ＋向ＣＤ４５ＲＯ＋细胞转化，而成人外周血淋巴细胞主要为ＣＤ３３－ＣＤ１６＋的ＮＫ细胞比例增加和ＣＤ４＋和ＣＤ８＋细胞亚群的ＣＤ２５＋细胞比例增加，而ＣＤ４５ＲＡ＋和ＣＤ４５ＲＯ＋细胞亚群无明显变化。

The flow cytometry was used to compare and analyse the effect of stimulation of rIL2 on the lymphoid subpopulations and its functions in human umbilical cord blood (UCB) and adult peripheral blood (APB).The results showed that the freshly isolated lymphocytes from UCB were immature in terms of phenotypes and in function compared with those from APB.The responsiveness to rIL2 was also significantly different between them.The changes of UCB lymphocytes were mainly in the CD4+ and CD8+ subsets,that was increase in the degrees of their maturation and the memory function or the subsets shifting from CD45RA+ to CD45RO+.The changes in APB lymphocytes were mainly increases of percentages of CD33-CD16+ NK cells in lymphocytes’ population and of CD25+ cells in CD4+ and CD8+ subsets.The changes of CD45RA+ and CD45RO+ were insignificant.The compositions and responsiveness to rIL2 of lymphocytes of both UCB and APB were different.
