摘要
目的观察金雀异黄素在低雌激素水平情况下对雌激素依赖阳性人乳腺癌细胞MCF-7的生长影响。方法采用四甲基偶氮噻唑蓝(MTT)法检测细胞增殖,流式细胞术分析细胞周期分布情况,磷脂结合蛋白V/碘化丙啶(AnnexinV/PI)联合双染法检测细胞凋亡,间接免疫荧光法测定雌激素调节蛋白(presenilin2,PS2)的表达。结果当MCF-7细胞经过去雌激素处理以后,与对照组相比,金雀异黄素在5×10^-7~10^-5mol/L时可以促进细胞增殖,G0/G1期细胞向S期推进,PS2蛋白表达增加,但是抑制细胞凋亡作用并不明显。结论金雀异黄素在低雌激素水平情况下可以促进人乳腺癌细胞MCF-7的细胞增殖、DNA合成以及PS2蛋白的表达,具有一定的雌激素样作用。
Objective To explore the effect of genistein on cell growth and possible mechanism of estrogen-dependent human breast cell MCF-7 at a low level of estrogen environment. Methods The proliferation effect of MCF-7 cell was analyzed by MTT. Flowcytometry and Annexin V/PI assay were used to analyze the cell cycle distribution and apoptosis. Indirect immunofluorescence assay was used to detect the expression of eestrogen-regulated protein ( presenilin 2, PS2 ). Results Compared with the control group,the cell proliferation, DNA synthesis and the expression of PS2 protein of MCF-7 cell were markedly enhanced by genistein at the concentration of 5 × 10^-7 --10^-5 mol/L. However, the effect of apoptosis inhibition was not obvious. Conclusion Genistein enhances cell proliferation, DNA synthesis and the expression of PS2 protein of human breast cancer cell MCF-7 at a low level of estrogen environment,which may demonstrate an estrogenic effect.
