摘要
目的采用HPLC—DAD中梯度洗脱和检测波长时间序列采样的方法,建立甘草(Glycyrrhiza uralensis Fisch.)中4种黄酮成分(甘草苷、异甘草苷、甘草素、异甘草素)和3种甘草皂苷成分(甘草酸、甘草皂苷G2、乌拉尔甘草皂苷B)的同时分析方法。方法采用C18(4.6mm×250mm,5um)色谱柱。流动相:乙腈-(0.5%醋酸铵+1%冰醋酸),梯度洗脱0~12min从6:94到22:78,12~25min从22:78到25:75,25—45min从25:75到40:60,45~50min从40:60到6:94,保持5min;检测波长:时间序列采样。结果7种成分的线性关系良好,平均加样回收率在98%~103%之间。结论该方法准确、灵敏度高,可用于甘草药材质量的评价。
OBJECTIVE To establish the method for simultaneous analysis of four flavone components ( liquiritin, isoliquiritin, liquiritigenin, isoliquiritigenin) and three saponin components (glycyrrhizin, licorice-saponin G2 and uralsaponin B) in Glycyrrhiza uralensis Fisch. using gradient elution and timetable detected wavelength by HPLC-DAD. METHODS The chromatographic separation was performed on a C18 (4. 6 mm×250 mm,5 um) column eluted with a mobile phase of aeetonitril-(0. 5% NH4Ac + 1% HAc ) , gradient program used was as follows :0 - 12 min 6:94 linear change to 22:78,12 -25 min 22:78 linear change to 25:75,25 - 45 rain 25:75 linear change to 40:60,45 -50 min 40:60 linear change to 6: 94, and this was maintained for 5 min and time-table detected wavelength. RESULTS Seven components had nice linear relation between the peak area and injected concentration. The average recoveries of the seven components were in the range from 98% to 103%. CONCLUSION The method was accurate, and sensitive. It's suitable to determine seven active components in G. uralensis and can be used for evaluating the quality of G. uralensis.
