线粒体细胞色素氧化酶亚基Ⅰ和Ⅱ mRNA的表达与精子活动力的相关性研究 被引量:1

摘要 目的比较线粒体细胞色素氧化酶亚基Ⅰ(COXⅠ)和Ⅱ(COXⅡ)在正常精子和弱精子症患者精子中的表达差异。方法对48份精液进行常规检查,收集活动力正常的精子和(a+b)级≤40%的弱精子症患者的精子,提取精子总RNA,采用反转录-聚合酶链反应(RT-PCR)方法检测COXⅠ,COXⅡ亚型mRNA的表达水平。结果与活动力正常的精子比较,COXⅠ和COXⅡmRNA在弱精子症患者精子中表达水平显著降低(P<0.05);精子密度与COXⅠ和COXⅡmRNA表达水平无相关性(r1=0.268,r2=0.042,P均>0.05)。结论人精子线粒体COXⅠ,COXⅡmRNA表达减少可能是精子活动力低下的原因之一。 Objective To compare the different expressions of mitochondrial cytochrome oxidase Ⅰ (COX Ⅰ ) and Ⅱ (COX Ⅱ ) in ejaculated spermatozoa between healthy volunteers and infertile males with asthenospermia (a+b ≤ 40%): Methods After routine examination of semen, spermatozoa of healthy volunteers and infertile males with asthenospermia (a+b ≤ 40%) were collected in the study. Total RNA of spermatozoa was extracted, and then mRNA expressions of COX I and COX II in spermatozoa were detected by RT-PCR. Results Compared with that in spermatozoa of healthy volunteers, mRNA expressions of COX Ⅰ and COX Ⅱ were all downregulated significantly in spermatozoa of patients with asthenospermia (P〈0.05). There was no relationship between spermatozoa density and levels of mRNA COX Ⅰ and COX Ⅱ (r1=0.268, r2=0.042, P〉0.05). Conclusion The down-regulated expression of COX Ⅰ and COX Ⅱ might be one reason for poor motivity of spermatozoa.
