摘要
目的研究LNCap和DU145细胞在体外培养时相关细胞特性及差异。方法常规培养LNCap和DU145细胞株。设6组:无血清空培养液组(A组)、有血精空培养液组(B组)、无血清培养LNCap组(C)组、有血清培养LNCap组(D组)、无血清培养DU145组(E组)和有血清培养DU145组(F组)。6组同时培养48h后,取各自培养液上清,经相关试剂盒ELISA分别检测睾酮(T)、雌二醇(E2)、孕酮(P)、血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、tPSA和fPSA浓度。结果T在A组未检出,B组因含血清而有少量T,在C、D和F组中T显示低浓度,但在E组却成倍升高。C、D、E和F组E2水平都高于对照组A、B组(P<0.05)。P在6组中差别都不大。VEGF在C、D、E和F组都有极其异常高表达(P<0.01),C、D组比对照组约高100倍,而E、F组比对照组则远高出1000倍。bFGF在C、D组与对照组差别不大,但在E、F组却显著升高(P<0.01)。E、F组中fPSA含量大于C、D组,在D、E和F组fPSA/tPSA均升高达44%。结论研究LNCap和DU145体外培养相关特性,可为我们进一步研究前列腺癌机制提供依据。
Objective To study biological features and related secreted proteins expression of prostate cancer cells LNCap and DU145 in vitro. Methods Cultured cells were divided intosix groups including: Blank culture without serum and without cell (group A), Blank culture with serum but without cell (group B), LNCap cell culture without serum (group C), LNCap cell culture with serum(group D), DU145 cell culture without serum (group E), DU145 cell culture with serum (group F). Total of 2 × 10^6 cells of LNCap or DU145 cells were seeded into each wellof 24-well-plate in Group C,D,E,F, for each plate-well. All test well were repeatedly done for three times. All the culture solution were collected after 48hrs culture..for the level of testosterone (T), estradiol (E2), progesterone (P), vascular endothelial growth factor(VEGF), basic fibroblast growth factor (bFGF), total prostate-specific antigen (tPSA) and free prostate specific antigen (fPSA) were detected with enzyme-linked immunoassay kits. Results The level of T was absent in group A,low in group B. middle in groups C,D,F, but much high in group E. The level of E2 was found robe high in groups C,D,E and F(P〈0.05). No distinct differences in level of P were found among all groups.There were extremely high expressions of VEGF in groups C,D,E and F(P〈0.01),and level of VEGF in groups C,D were higher 100 folds and 1000 times than groups A,B. The level of bFGF were distinctly higher in groups E,F than that of groups A,B,C and D(P〈0.01). The level of fPSA in groups E,F were higher than that in groups C,D, and the ration of fPSA/tPSA all increased in groups D,E and F. Conclusion Although these two cell lines derived from same resource of human distinct differences in biological characteristics were still found between cell LNCap and DU 145 cell lines, which might be helpful for elucidating the mechnism of Prostate Cancer.
