尾加压素Ⅱ及其受体在2型糖尿病小鼠肝脏中表达的变化 被引量:4

摘要 目的观察2型糖尿病(type 2 diabetes mellitus,2DM)小鼠肝脏中尾加压素Ⅱ(urotensinⅡ,UⅡ)及其受体表达的变化。方法应用血糖测定仪检测小鼠空腹血糖浓度,应用放射免疫方法检测血浆中UⅡ及胰岛素的含量,用免疫组化方法观察UⅡ在肝脏中的表达,应用RT-PCR法测定GPR14 mRNA在肝脏的表达。结果2DM小鼠空腹血糖及血浆胰岛素含量高于正常对照组,血浆UⅡ含量高于正常对照组,肝脏免疫活性UⅡ表达及其受体GPR14 mRNA的表达均明显高于正常对照小鼠。结论2 DM小鼠肝脏UⅡ表达增高可能是血浆UⅡ增高的来源之一,受体GPR14 mRNA表达也增高,其在2DM发病中的意义值得进一步探讨。 Objective To study the expression changes of UⅡ and its receptor in 2DM mouse liver. Methods Blood glucose monitoring device was used to examine the concentration of fast blood glucose. Radioimmunity assay method was used to examine the concentration of plasma UⅡ and insulin. RT-PCR method was used to assay the expression of GPR14 mRNA in mouse liver. Immunohistochemistry method was used to assay UⅡ immunological activity expression in mouse liver. Results Compared with control group, the concentration of fasting blood glucose, plasma insulin and plasma UⅡ increased in 2DM group, immunological activity of UⅡ was strong positive expression in 2DM mouse liver, the expression of GPRI4 mRNA was up-regulated in 2DM mouse liver. Conclusion It is possible that the increasing expression of UⅡ derived from liver is a source of plasma UⅡ. But it is unclear what relevance the upregulated expression of GPR14 mRNA has with 2DM and deserves further study.
