热休克蛋白70单抗诱导胰腺癌细胞凋亡的实验研究 被引量:2

摘要 目的探讨HSP70单抗对人胰腺癌细胞株BXPC-3的生长抑制作用和凋亡的诱导作用及可能的作用机制。方法MTT法检测HSP70单抗对人胰腺癌细胞株BXPC-3增殖的影响,流式细胞仪检测HSP70单抗诱导凋亡作用。结果MTT法检测24h,48h不同浓度HSP70单抗作用后实验组与对照组吸光度(A)值比较差异有统计学意义(P<0.05),48h的抑制率高于24h的抑制率。流式细胞仪分析显示HSP70单抗作用48h后细胞发生凋亡,且随单抗浓度的增加细胞凋亡率增高。结论HSP70单抗可以抑制BXPC-3细胞的增殖并且可以诱导其凋亡。 Objective To investigate the effect of HSP70 monoclonal antibody on the growth of human pancreatic cancer cell line BXPC-3 inhibition and induced apoptosis and the possible mechanism. Methods MTT assay was used to detect the proliferation effect of HSP70 monoclonal antibody on human pancreatic cancer cell line BXPC-3. Flow cytometry was used to detect the apoptosis induced by HSP70 monoclonal antibody. Results MTT assay showed that there was a significant difference in absorhance a values ( P 〈 0.05 ) between different concentrations of HSP70 monoclonal antibody experimental group and control group 24 h, and 48 h later. The Inhibition rate in 48h was higher than 24 b. Flow cytometry showed that the role of HSP70 monoclonal antibody after 48 h cell apoptosis, and the rate of apoptosis was increased with the monoclonal antibody concentration increasing. Conclusion HSP70 monoclonal antibody can inhibit the proliferation of BXPC-3 cell and induced apoptosis.
