益智通脉颗粒对血管性痴呆大鼠脑组织VEGF表达的影响

The Effects of Yizhitongmai Granule on VEGF of Cerebral Tissue of Vascular Dementia in Rats

目的探讨益智通脉颗粒对血管性痴呆大鼠脑组织VEGF表达的影响。方法健康成年雄性Wister大鼠50只随机分为正常对照组、模型对照组、中药大剂量组、中药小剂量组、尼莫地平组各10只,采用双血管阻断法制备拟血管痴呆大鼠模型。术后7d模型组及药物组分别给药,连续给药60d。采用免疫组化法观察脑组织VEGF的表达。结果中药大、小剂量组与模型组及尼莫地平组比较有显著差异;中药大、小剂量组之间呈现正相关量-效关系。结论益智通脉颗粒可通过上调VEGF表达,促进内皮细胞增殖及毛细血管新生,从而起到脑保护作用。

Objective： To observe the effects of Yizhitongmai Granule on VEGF of cerebral tissue of vascular dementia（VD）model rats. Methods： 50 male Wistar rats were randomly divided into 5 groups： the normal group,the model group, high - dose group （treated with high dose Yizhitongmai Granule）, low - dose group and nimodipine group. Rat VD model was established by the method of both of vessel -blocked, and then 7d later, treating the rats with medicine for 60d. SP immunohistochemical staining was used to examine expressions of VEGF of cerebral tissue. Results： There were significantly difference by high -dose and low- dose group comparing with model group and nimodipine group. There were positive correlation between high-dose and low-dose group. Conclusion： Yizhitongmai Granule can facilitate proliferation of endothelial cell, accelerate vascularization and increase vasucular permeability by upregulate VEGF expression to protection brain.
