摘要
目的探讨丹参及其复方对急、慢性肺损伤(ALI/CLI)大鼠肺水通道蛋白-1(AQP1)等指标的影响。方法SD大鼠随机分为ALI组(30只)与CLI组(50只),ALI组随机分为正常组、治疗组、模型组各10只;CLI组随机分为模型组、丹参复方治疗组、西药美吡达治疗组、糖尿病组及正常组各10只;造模及给药后各组分别行血液流变学检测,电镜及HE染色观察,肺湿/干重比(W/D),免疫组化及Western Blot法检测AQP1表达变化。结果各模型组均出现肺损伤症状。丹参治疗组与ALI模型组比较肺水肿病理改变明显减轻,血液流变学指标明显改善。DM大鼠肺泡间隔及毛细血管壁增厚,AQP1表达下降,丹参复方组较模型组其它各组表达升高。结论急、慢性肺损伤时丹参及其复方均能增加肺毛细血管AQP1的表达,改善血液流变性,减轻肺组织病理改变,在一定程度上缓解肺损伤病变的发生与发展。
Objective: To investigate the effects of salvia mihiorrhiza(SM) and its decoction on aquaporin - 1 (AQP1) expression in rats with acute lung injury(ALI) and chronic lung injury(CLI) . Methods: 80 SD rats were divided into ALI group(30) and CLI group(50). ALI group randomly divided into normal group, ALl group and SM prevention group, 10 rats per group. CLI group randomly divided into SM decoction group, minidiab treatment group, model group and normol group. After treatment, lung wet- to- dry weight ratio, lung pathology, electronmicroscope, AQP1 immunohistochemistry and AQP1 Western Blot were performed. Results: All model group rats appeared ALI and CLI symptoms. Compared with ALl group, the degree of lung edema statement alleviated in SM treatment group. Compared with DM group, the pathology changes of lung alleviated in SM decoction group. The expression of AQP1 was significantly expressional in both the group of SM prevention group and SM decoction group. Conclusion: SM and its decoction may improve the body fluid metabolism, the situation of hemorheology and alleviate the degree of the lung edema by regulating the expression of AQP1.
