益智宁煎剂对SHR大鼠前额叶皮质及纹状体多巴胺的影响 被引量:5

摘要 目的观察中药复方益智宁煎剂对注意缺陷多动障碍(ADHD)动物模型自发性高血压大鼠(SHR)前额叶皮质及纹状体多巴胺(DA)的影响。方法将11周龄的雄性SHR分为益智宁高剂量组、低剂量组和生理盐水对照组,分别予益智宁和生理盐水灌胃,用高效液相色谱—电化学法检测前额叶皮质、纹状体中多巴胺的含量。结果益智宁可明显升高SHR纹状体多巴胺含量,高剂量组、低剂量组与生理盐水对照组比较差异有统计意义(P<0.05),但高、低剂量组间无明显差异;3组间前额叶皮质中多巴胺的含量比较,差异无统计意义(P>0.05)。结论益智宁治疗肾虚肝亢型ADHD的作用机制之一可能与调节单胺类神经递质有关,其对额—纹状体环路DA的调控主要在于提高纹状体DA含量,纹状体为作用靶点之一。 Objective To study the influence on the dopamine (DA) in frontal lobe cortex and striate body of spontaneity hypertensive rats (SHR) model of attention deficit hyperactivity disorder (ADHD) by Yizhining Decoction. Methods 11-weeks male SHR were chosen to be the animal models, divided into highdose Yizhining group, low-dose Yizhining group and normal sodium-control group. SHR were given corre- sponding drugs orally. Dopamine levels in frontal lobe cortex and striate body were detected by HPLT-electrochemical method. Results Compared with normal sodium-control group, high-dose and low-dose Yizhining groups could obviously increased the dopamine level in striate body (P 〈 0.05). But there was no significant difference between the two treatment groups. Meanwhile, there had no difference in the high-dose Yizhining group, low-dose Yizhining group and normal sodium-control group in frontal lobe cortex ( P 〉 0.05 ). Conclusion The possible mechanism of Yizhining Decoction on kidney-deficiency and liver-hyperactivity in ADHD may be related to that it can increase the dopamine level in striate body.
