摘要
目的通过存活素(Survivin)反义寡核苷酸(ASODN)转染下调人卵巢癌细胞株SKOV3中Survivin蛋白表达,观察细胞侵袭能力的改变,探讨Survivin蛋白表达可能在调节卵巢癌细胞侵袭能力中的作用。方法用脂质体(LipofectamineTM2000)介导ASODN转染人卵巢癌细胞株SKOV3,流式细胞学技术检测细胞转染率及其Survivin蛋白表达,Transwell小室检测细胞侵袭能力的改变。结果LipofectamineTM2000介导ASODN人卵巢癌细胞株SKOV3转染,FCM检测转染率>95%。用600 nm/L ASODN转染48 h后,细胞中Survivin蛋白表达明显下调(t=7.8146,P<0.01),侵袭细胞数也明显下降(t=27.416,P<0.01)。结论卵巢癌细胞中Survivin蛋白表达下调,可降低卵巢癌细胞侵袭能力,提示Survivin蛋白可能是影响卵巢癌侵袭能力的重要分子,在卵巢癌转移机制中起关键性作用。
Objective To explore the effects of down-regulation of Survivin protein on the invasion in ovarian cancer cell line (SKOV3) by transfeeting anti-survivin oligonueleotides (survivinASODN). Methods SKOV3 cells were transfeeted with anti-Survivin oligonueleotides (ASODN) by LipofeetamineTM2000 transfection reagent, and the rate of transfection were determined by FCM. The changes of Survivn protein were detected by flow eytometry (FCM), and the invasion ability was detected by Transwell. Results Transfeetion rate of ASODN in SKOV3 cells was above 95 %. The protein levels of Survivin was significantly down-regulated after 48 hours of transfeetion with 600 nm/L ASODN, ( t = 7.8146, P 〈0.01), The number of invasive cells was also reduced ( t = 27.416, P 〈0.01). Conclusion Down regulation of Survivin protein can reduce the invasive ability of ovarian cancer cells. Survivin could be a crucial factor in regulating invasion ability of ovarian cancer cells, and may play an important role in the molecular mechanism of the metastases of ovarian cancer.
