Ca^(2+)和PKc在巨噬细胞激活过程中作用的初步研究

摘要 ①目的探讨Ca2+和蛋白激酶C(PKc)在巨噬细胞激活过程中的作用。②方法在不同试验条件下,用同位素标记的方法,检测巨噬细胞介导的细胞毒作用及PKc的活性。③结果胞外Ca2+络合剂不影响巨噬细胞活化因子(MAF)的作用,而钙离子通道剂A23187激活巨噬细胞的作用完全丧失;胞内Ca2+拮抗剂能够抑制MAF或A23187激活巨噬细胞的作用,并且有剂量依赖关系;巨噬细胞激活剂能使胞质中PKc活性大大降低,而使膜结合PKc活性明显升高。 Objective To study the function of intracellular second signals in macrophage activation. Methods Macrophage mediated cytotoxicity and protein kinase C(PKc) activity under different conditions were measured by isotope labelling method. Results Extracellular calcium antagonists blocked the stimulating effect of calcium ionophore A 23187 ,but exerted no effect on MAF; Macrophage activation by MAF or A 23187 was inhibited by intracellular calcium antagonists,and the inhibition effect was concentration dependent; PKc activity in cytosol was greatly decreased by macrophage priming agents, but PKc in cell membrane was elevated significantly. Conclusion Ca 2+ and PKc had important effect on signal transportation in macrophage activation.
