摘要
采用花粉管通道法将含有Bt杀虫基因的pG4AB质粒导入受体棉花中23中,通过卡那霉素筛选,Bt免疫检测条检测,PCR扩增对转基因抗虫棉的各个自交世代进行跟踪研究。结果表明,具有卡那霉素抗性的单株能扩增出Bt基因,而且均有Bt杀虫蛋白表达。Bt基因与npt Ⅱ基因均整合进了棉花基因组中,相互之间紧密连锁遗传,均稳定表达,因此报告基因npt Ⅱ可用于研究Bt基因的遗传情况。
Plasmid pG4AB containing Bt gene was transferred into cotton Zhong 23 via pollon tube pathway. The Kanamycin detection and the expression of Bt ICP, PCR were used to analyze the self-pollination generations from transgenie cotton Zhong 23. The results were as follows. Exogenous Bt gene was amplified and expressed in the Kanamyoin-resistance transgenic cotton. The Bt gene and npt Ⅱ gene were inserted into the transgenic cotton genome, they were linked and inherited stably in different generations, npt Ⅱ gone can be used in inheritance analysis of Bt gene.
