籼稻扬稻6号和粳稻苏沪香粳花药培养力的比较 被引量:6

摘要 为建立适合江苏水稻主栽品种花药培养的技术体系,本研究以籼稻扬稻6号和粳稻苏沪香粳为研究材料,分别从取样温度,低温预处理天数以及培养基中的蔗糖、琼脂和激素等研究不同水稻亚种花培的植株再生技术体系.结果表明:在气温26℃~31℃之间,扬稻6号在4℃低温预处理9d,接种到以M8为基本培养基,附加2,4-D1mg/L,NAA3mg/L,KT1mg/L,脯氨酸100mg/L,水解酪蛋白300mg/L,60g/L蔗糖和琼脂8g/L的花药愈伤诱导培养基中,可以获得高达8.5%的出愈率.而苏沪香粳在气温27℃~30℃之间,4℃低温预处理12d,接种到以M8为基本培养基,附加2,4-D1mg/L,NAA3mg/L,KT1mg/L,脯氨酸100mg/L,水解酪蛋白300mg/L,60g/L蔗糖和琼脂6g/L的花药愈伤诱导培养基中,可以获得高达15.05%的出愈率.扬稻6号和苏沪香粳花培愈伤组织转接分化培养基分别获得2.7%和5.7%的绿苗率. The culture ability of callus from anther of two rice varieties, Indica Rice Yangdao 6 and Japonica Rice Suhuxiangjing are studied as materials. The results show that when using M8 as basic induction medium adding 60 g/L sucrose, 6 g/L agar, 1 mg/L 2,4 - D, 3 mg/L NAA, 1 mg/L KT, 100 mg/L proline and 300 mg/L hydrolyzed casein protein, inductivity of callus in Yangdao 6 is high as 8.5 percents after chilling treatment for 9d during 26℃ - 31℃ air temperature for sampling. While that in Suhuxiangjing is high as 15.05 percents when using M8 as basic medium adding 60 g/L sucrose, 8 g/L agar, 1 mg/L 2,4 - D, 3 mg/L NAA, 1 mg/L KT, 100 mg/L proline and 300 mg/L hydrolyzed casein protein; after chilling treatment for 12 d during 27℃ -30℃air temperature for sampling. And that when using MS as differentiation basic medium adding 0. 2 mg/L 2,4 - D ,2 mg/L 6 - BA ,2 mg/L KT,the frequency of regeneration of callus from anher in Yangdao 6 and Suhuxiangjing are 2. 7 percents and 5.7 percents, respectively. The stable and effective regeneration from anther in Yangdao 6 and Suhuxiangjing are established.
