摘要
采用DEAE-52纤维素柱色谱和Sephadex G-100凝胶柱色谱对苦瓜多糖(MCP)进行分离纯化研究。利用高通量筛选法对苦瓜多糖的分离纯化产物进行降血糖降血脂活性研究。试验以SMMC-7721肝癌细胞为模型,确定了苦瓜多糖各个样品的合适浓度为50μg/mL,并以此浓度对PPARγ、PPARδ受体的激活能力进行了筛选。结果表明,苦瓜多糖分离产物MCP2和MCP4对PPARγ受体有激活作用,激活倍分别为1.726和1.602。MCP2-1和MCP2-2样品都能激活PPARγ受体,激活倍分别为1.593、1.678。MCP2、MCP2-1和MCP2-2是苦瓜多糖的降糖活性成分。
Momordica charantin linn polysaccharides (MCP) was isolated by DEAE ion-exchange cellulose (DE-52) and Sephadex G-100 gel filtra- tions. Screening anti-diabetes and anti-hyper activity of MCP with high throughput screening (HTS) was studied in this paper. The SMMC-7721 cells were employed as model cell. The best concentration of each extracts was 50 μg/mL, and screening PPARγ, PPARδ agonist from momordica charantia extracts by HTS with this concentration. Results showed that MCP2 and MCP4 were agonists of PPARγ, and the activity times were 1.726 and 1. 602, respectively. MCP2-1 and MCP2-2 were agonists of PPAR2,, and the activity times were 1.593, 1. 678, respectively. MCP2, MCP2-1 and MCP2-2 could be potential anti-diabetie agents.
