利用染色体片段置换系定位水稻酚反应基因 被引量:3

Mapping of QTLs for Phenol Reaction Using Chromosome Segment Substitution Lines in Rice(Oryza sativa L.)

摘要 水稻谷粒苯酚染色反应是鉴别籼粳稻亚种的指标之一。本研究以9311和日本晴构建的8个染色体片段置换系为材料,采用代换作图法对控制水稻谷粒酚反应的3个QTL进行了定位。结果表明,4个染色体片段置换系材料酚反应级别为4,其余的在0到1级。8个置换系均含有来源于日本晴的1个置换片段,其中3个置换系在第2染色体、2个置换系在第4染色体和3个置换系在第7染色体上分别有1个置换片段,其长度分别为24.9cM、37.8 cM、9.7 cM、9.5 cM、22.7 cM、16.7 cM、22.2 cM和13.0 cM,平均长度为19.6 cM。3个控制水稻苯酚反应的基因qPH4-1、qPH2和qPH7被界定在第2染色体RM406和RM240、第4染色体RM280和RM5709及第7染色体RM11和RM6574之间,遗传距离分别为12.9 cM、9.5 cM和16.0 cM。qPH4-1、qPH2和qPH7的鉴定和初步定位为其进一步的精细定位和图位克隆奠定了基础。 The phenol reaction of rice seed is one of the indexes differentiating japonica from indica. In this paper, chromosome segment substitution lines( CSSLs), derived from 9311×Nipponbare with 9311 as the recurrent parent,were experimented for the phenol reaction, and three QTLs were analyzed by substitution mapping. The results showed that four chromosome segment substitution lines (CSSLs) were class 4 and the rest were 0 to 1 class. All the CSSLs had one introgressed segment coming from Nipponbare,three CSSLs on rice chromosome 2 ,two CSSLs on rice chromosome 4 and three CSSLs on rice chromosome 7 having one introgressed segments, respectively. The genetic distances of the eight introgressed segments were 24. 9 cM,37.8 cM,9.7 cM, 9.5 cM,22. 7 cM,16.7 cM,22.2 cM and 13.0 cM,respectively. Three QTLs for the phenol reaction were identified, qPH2 was mapped in the region of RM406 and RM240,which was 12. 9 cM in genetic distance on rice chromosome 2. qPH4-1 was mapped in the region of RM280 and RM5709, which was 9.5 cM in genetic distance on rice chromosome 4. qPH7 was mapped in the region of RMll and RM6574,which was 16. 0 cM in genetic distance on rice chromosome 7. The QTLs for the phenol reaction of rice seed can be mapped primely using chromosome segment substitution lines. Mapping of the QTL has laid a foundation for fine mapping and cloning of the QTL in rice.
