摘要
本研究在已获得产气荚膜梭菌β2-β1毒素融合基因表达载体,并在BL21(DE3)中成功表达的基础上,研究了以乳糖为诱导剂在不同条件下对β2-β1重组蛋白诱导表达的影响,并对其表达条件进行了优化。结果表明,重组菌株BL21(DE3)(pXETB2B1)在37℃、菌体诱导密度OD600为1.0、乳糖诱导浓度为0.1 g/L时诱导5 h,目的蛋白的表达效果最好,乳糖则以分批添加的方式为佳。
Based on the expression of β1 toxin and β2 toxin fusion gene of Clostridium perfrigens in the recombinant strain BL21 (DE3) (pXETB2B1) ,the expression condition of the fusion protein was optimized with lactose as inducer. The highest expression was obtained when the concentration of lactose was 0. 1 g/L,induction time was 5 h, cell density of induction was OD_600 1.0 and the temperature was 37℃,respectively. The best way to add lactose was the batch processing.
