摘要
目的探讨H2S对大鼠肝星形细胞(HSC)增殖和氧应激的影响。方法用铁超载的方法复制HSC增殖模型,通过绘制生长曲线以及CCK-8细胞计数法检测不同浓度的H2S对500μmol/L次氮基三乙酸铁(FeNTA)作用下的HSC增殖的影响。通过不同浓度的H2S作用于500μmol/L FeNTA培养的HSC,检测6h、12h、24h细胞上清液和裂解液中H2S、SOD活力、MDA含量,观察H2S对HSC氧应激反应的影响。结果同对照组比较,一定浓度的H2S对HSC增殖有抑制作用。在FeNTA诱导的氧应激反应中,一定浓度范围的H2S可有效提高SOD的活性和降低MDA含量。结论H2S对HSC增殖有较强抑制作用,对FeNTA诱导的氧应激具有保护作用。
Objective To investigate the effects of oxidative stress on the proliferation of rat hepatic stellate cells (HSC). Methods HSC were incubated with FeNTA. The effect of various concentrations of H2S on the proliferation of rat HSC incubated with 500 mol/L FeNTA were measured by making cell growth curves and CCK - 8 counting. After rat HSC were incubated with various concentrations of H2S and FeNTA for 6h, 12h, and 24h, the activity of superoxide dismutase (SOD) and contents of H2S and malondialdehyde (MDA) in culture supernatant and cell lysis solution were detected to observe the effect of H2S on the oxidative stress of rat HSC. Results Compared with the control group, the proliferation of rat HSC was inhibited when the concentration of HES in the medium reached to a certain range . In the oxidative stress induced by FeNTA, H2S within certain strength range, obviously enhanced the activity of SOD and decreased the contents of MDA in supernatants of rat HSC culture media. Conclusions H2S could inhibit the proliferation of rat HSC and it, within a certain strength range, exert protective effect in oxidative stress induced by FeNTA.
