摘要
目的探讨EEF1A2转基因对胰腺癌细胞SW1990生长和增殖的作用。方法应用腺病毒载体将EEFIA2基因转染人胰腺癌细胞SW1990,采用MTY法检测细胞的增殖,软琼脂克隆形成试验检测细胞的生长,应用流式细胞仪检测细胞周期的变化。结果带EEFIA2的腺病毒感染SW1990细胞后,EEF1A2mRNA表达增加,72h的A750值为1.2996±0.2091,培养6d的细胞数为81250±1767,14d的克隆形成率为82%,均较空载体腺病毒组和PBS组显著增加(P〈0.05);G.期细胞比例为28.5%,S期细胞比例为60.9%,前者较空载体腺病毒组和PBS组显著减少,后者较空载体腺病毒组和PBS组显著增加(P〈0.05)。结论EEF1A2基因可以显著促进人胰腺癌细胞SW1990的生长和增殖。
Objective To investigate the effects of EEF1A2 on growth and proliferation of the human pancreatic cancer cell line SW1990. Methods The EEF1A2 gene was introduced into pancreatic cancer cell line SW1990 by adenovirus vector. The effects of EEF1A2 on the growth of human pancreatic cancer cell were measured by MTF. Cell cycle was detected by flow cytometry and cell growth rate was examined by soft agar cloning formation test. Results After EEFIA2 induction, the expression of EEFA1A 2 mRNA in pancreatic cancer cell line SW1990 increased, value of A750 at 72 h was 1. 2996±0. 2091, number of cells was 81 250, cloning efficiency at 14 d was 82%, all of these parameters were significantly higher than those in the groups of blank adenovirus vector and PBS groups ( P 〈 O. 05 ) ; the percentage of G1 phase cell was 28.5%, which was significantly lower than those in the groups of blank adenovirus vector and PBS groups; the percentage of S phase cell was 60.9%, which was significantly higher than those in the groups of blank adenovirus vector and PBS groups (P 〈 0.05). Conclusions EEF1A2 gene significantly enhanced cell growth and proliferation in human pancreatic cancer in vitro.
