摘要
目的:观察桂枝芍药知母汤对免疫性关节炎大鼠血清中TNF-α与关节滑膜组织Bc l-2表达的影响,并初步探讨其治疗机制。方法:以弗氏完全佐剂诱导产生免疫性关节炎模型,用不同剂量桂枝芍药知母汤(GSZ汤20.6、10.3 g/kg)给模型动物灌胃用药,观察用药后动物血清TNF-α水平与致炎对侧膝关节滑膜组织Bc l-2表达。结果:桂枝芍药知母汤用药4w后,可明显使关节指数减小(P<0.05或P<0.01);使模型大鼠血清TNF-α水平降低(P<0.01);免疫组化染色镜检显示,大鼠膝关节滑膜组织Bc l-2阳性细胞数减少、阳性细胞着色强度减弱(P<0.05或P<0.01),本实验条件下,GSZ汤20.6 g/kg剂量时的疗效与MTX相当。结论:桂枝芍药知母汤对免疫性关节炎大鼠有治疗作用,其机制可能与抑制TNF-α分泌,降低Bc l-2表达有关。
Objective :To observe the therapeutic effect of guizhi-shaoyao-zhimu (GSZ)decoction on immunologic arthritis rats and investigate the mechanism by determining the expression of serum TNF-α and Bcl-2 in synovial tissues of joint. Methods:The rats were prepared for immunologic arthritis models by Freund's complete adjuvant(FCA). Different doses of GSZ decoction (20. 6 g/kg, 10. 3 g/kg) were administrated on rat models. The serum of experimental animals were collected to detect TNF-α, and the counter-lateral synovial tissues of the knee joint were taken for observing the expression of Bcl-2 by immunohistochemical test. Results : In a 4-week administration of GSZ decoction,lessened arthritis index was observed (P 〈 0. 05 or P 〈 0. 01 ) ,which resulted in a lower serum TNF-α in the model rats ( P 〈 0. 01 ) ;The immunohistochemical test suggested Bcl-2 positive staining cell count became smaller and weakened (P 〈0. 05 or P 〈0.01). Moreover,the findings suggested that GSZ decoction 20. 6 g/kg could be favorable with respect to MTX in treatment of immunologic arthritis rats. Conclusion: GSZ decoction can treat the joint impairment of immunologic arthritis rats. The therapeutic mechanism may be relevant to reducing the content of TNF-α and inhibiting the expression of Bcl-2.
