槲皮素-3-葡萄糖苷对小鼠心肌缺血缺氧损伤的保护作用 被引量:14

摘要 目的观察槲皮素-3-葡萄糖苷(QG)对小鼠心肌缺血缺氧的保护作用。方法采用夹闭气管造成小鼠心肌缺氧模型,观察QG对气管夹闭小鼠心电持续时间的影响;采用皮下注射异丙肾上腺素致小鼠急性心肌缺血模型,观察心肌缺血后心电图ST段的改变、心肌组织中乳酸脱氢酶(LDH)活力的变化,同时观察心肌组织中丙二醛(MDA)的含量、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-PX)活力的变化。结果5、10mg/kgQG能明显延长气管夹闭小鼠的心电持续时间,抑制异丙肾上腺素造成的心电图ST段的抬高,升高心肌组织中LDH及GSH-PX的活性,并同时降低心肌组织中MDA的含量。2.5、5、10mg/kgQG能明显升高心肌组织中SOD活性。结论QG对小鼠心肌缺血缺氧具有一定的保护作用,其作用可能与提高机体抗氧自由基脂质过氧化有关。 Objective To study protective effects of quercitrin-3'-glucoside (QG)on acute myocardial ischemia in mice. Methods The duration of electrocardiograph (ECG) after clamping trachea was observed. Acute myocardial ischemia model was induced by subcutaneous injection of isoprenaline (Iso). The change of ST segment of ECG was detected. The activities of lactate dehydrogenase (LDH), superoxide dismutase (SOD), glutathione perioxidase (GSH-PX) ,and the content of maleic dialdehyde (MDA) in myocardium were measured. Results 5 and 10 mg/kg QG markedly prolonged the duration of ECG during clamping trachea in mice, inhibited Iso-induced the elevation of ST segment of ECG; 5 and 10 mg/kg QG obviously inhibited the reductions of LDH and GSH-PX activities, and decreased content of MDA in myocardium. 2.5, 5 and 10 mg/kg QG significantly inhibited Iso-induced the reduction of SOD activity. Conclusion QG has protective effects on mice myocardial ischemia via attenuating oxygen free radicals.
