摘要 目的获得酸性海藻糖酶基因(ATM1)并对其序列进行分析。方法根据已知序列设计引物,PCR扩增出金龟子绿僵菌ATM1的cDNA和DNA序列,对其推导的氨基酸序列基于多序列局部比对和结构的序列比对进行分析。结果ATM1cDNA序列包含开放阅读框(ORF)3219bp,3’端非翻译区(3’UTR)175bp和5’端非翻译区(5’UTR)498bp。结论成功获得ATM1基因,通过序列分析确定出4组保守性功能序列。ATM1基因在CQMal02基因组中为单拷贝。 Objective To obtain and anlyse the gene ATM1. Methods On the basis of a partial ATM1 sequence obtained, the whole cDNA sequence and its whole genomie DNA of ATM1 gene in M. anisopliae CQMa102 were amplified by using 3'race and 5'race. ATM1 gene was analysed based on sequence local alignment and structure-based sequence alignment. Results ATM1 gene has three introns and its ORF encodes a 1 073 amino acid protein. Conclusion Four sequence motifs of ATM1 have been indentified. By southern analysis it is indicated that ATM1 gene was present as a single copy in this M. anisopliae strain.
