白藜芦醇诱导前列腺癌细胞系PC-3凋亡的实验研究 被引量:5

摘要 目的探讨白藜芦醇(resveratroi,Res)诱导人前列腺癌细胞系PC-3凋亡及其机制。方法应用四甲基偶氮唑蓝(MTT)比色法、软琼脂集落生长试验和流式细胞术测定Res对PC-3的增殖抑制、集落生长抑制、周期阻滞和诱导凋亡作用,应用逆转录-聚合酶链反应(RT-PCR)方法检测对凋亡相关基因bcl-2和bax表达的影响。结果Res具有时间和剂量依赖性增殖抑制、G0/G1期阻滞和诱导凋亡作用,作用72 h IC50为0.39 g/L,Res 0.1 g/L作用2天对PC-3细胞集落生长抑制率为88.9%,Res 0.5 g/L作用2天G0/G1期细胞比例从42.30%增高至75.20%。Res可以下调bcl-2基因/蛋白,上调bax的表达。结论Res可能通过抑制PC-3细胞的集落生长、增殖抑制、G0/G1期阻滞、下调bcl-2基因,上调bax基因表达等作用机制诱导前列腺癌细胞凋亡。 Objective To investigate the effects of Res on prostate cancer cell line PC-3 in vitro . Methods The effects of Res on anti-proliferrafion, inhibition of clonal growth, cell cycle arrest and apoptosis induction were observed by MTT assay, colony on soft agar and flow-cytometry;RT-PCR method was used to examine the expression level of apoptosis-related genes bcl-2, bax. Results Res could cause apparent anti-proliferation effect, induce G0/G1 phase arrest and apoptosis induction time-dependently and dose-dependently on PC-3 cell. Inhibition of cell growth by 50% ( IC50 ) was seen at concentration of Res 0.39 g/L. The clonal growth inhibition rate was 87.9% after PC-3 cells treated by Res 0.1 g/L for 2 days;The ratio of G0/G1 phase cell raised from 42.30% to 75, 20% after treatment with Res 0.6 g/L for 2 days. Res also decreased the expression of bcl-2 gene,increased the level of bax genes. Conclusion It seems that Res has the antitumor action through anti-proliferation, inhibition of clonal growth, G0/G1 phase arrest, apoptosis induction, decreasing bcl-2 and increasing bax.
