PCR-SSCP法检测人胃癌组织中Rhobtb2基因突变的研究

摘要 目的观察Rhobtb2基因在人胃癌组织中的突变情况,探讨Rhobtb2基因突变与胃癌发生的关系。方法采用聚合酶链式反应-单链构象多态性(PCR-SSCP)方法对58例胃癌组织以及癌旁正常组织的Rhobtb2基因进行突变检测,采集1例正常人抗凝血作为正常对照。结果58例胃癌组织中发现1例位于外显子5的错义突变。结论胃癌组织中存在Rhobtb2基因突变,其突变可能与胃癌的发生有关。 Objective To investigate the mutation of Rhobtb2 gene and explore the relationship between Rhobtb2 mutation and sporadic gastric cancer. Methods Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was used to detect exons of Rhobtb2 from 58 patients with gastric cancer. The anticoagulant blood was collected from one normal person as normal control group. Results There was one case of missense mutation in exon5 in 58 cases of gastric cancer tissues. Conclusion There was Rbobtb2 mutation in sporadic gastric cancer, and the mutation may be associated with the tumorigenesis of gastric carcinoma.
