热带假丝酵母菌ERG11基因突变与氟康唑耐药性的关系 被引量:2

摘要 目的探讨热带假丝酵母菌氟康唑作用的靶酶编码基因(ERG11)突变与耐药性的关系。方法根据ERG11序列设计引物,对氟康唑靶酶基因ERG11进行PCR扩增,DNA测序后对比分析耐药株与敏感株及标准株的碱基差异,确定耐药株基因一级结构上的突变点,并推测分析靶酶蛋白的氨基酸改变及与氟康唑耐药的关系。结果氟康唑敏感株的ERG11基因DNA序列与氟康唑敏感标准株ATCC750完全一致,耐药株出现+395、+513、+783处点突变,导致132位氨基酸改变。结论ERG11基因突变所编码的氨基酸改变与氟康唑耐药有关。 Objective To explore the relationship between ERG11 gene mutation and fluconazole-resistance of Candida tropicalis. Methods ERG11 genes of Candida tropicalis 'were amplified by PCR, and their nucleotide sequences were determined in order to detect point mutations related to fluconazole-resistance. Results The results showed 3 base mutations at +395, +513, +783 positions, which could result a single amino acid substitution in the fluconazole-resistance strain. Conclusion It is suggested that the gene mutation of ERG11 on the amino acid sequence is relative to the fluconazole-resistance.
