摘要
目的研究白花蛇舌草乙酸乙酯提取液对多药耐药白血病细胞HL-60/ADR的抑制作用及其机制。方法采用MTT比色试验观察白花蛇舌草乙酸乙酯提取液对多药耐药细胞HL-60/ADR的抑制作用。采用光镜、电镜技术观察细胞形态结构的改变,利用流式细胞术检测细胞凋亡率。结果①白花蛇舌草乙酸乙酯提取液能明显抑制HL-60/ADR细胞的生长,药物浓度相当于生药41.67mg/ml时,即具有显著的抑制作用,抑制率呈剂量和时间依赖性,半数抑制浓度(IC50)相当于生药70.41mg/ml。②形态学改变,呈典型的凋亡特征。③白花蛇舌草乙酸乙酯提取液能诱导多药耐药细胞HL-60/ADR凋亡,凋亡率也呈时间和剂量的依赖性。结论白花蛇舌草对多药耐药细胞HL-60/ADR的生长具有极强的抑制作用,诱导其凋亡是其机制之一。
Objective To approach into the inhibitory action and mechanism of acetic acid ethyl ester extracted from Hedyotie diffusa on leukemic cell line HL-60/ADR with multidrug resistance. Methods The depressant effect on multidrug resistance leukemic cell line HL-60/ADR were observed by MTT. The change of cellular morphology were analysed under light microscope and electron microscope. The apoptosis rates were detected by flow eytometry. Results i The extracts from Hedyotic diffusa could effectively inhibit the leukemic cell proliferation, ii Some of the cells formed apoptotic bodies with typical characters, iii Acetic acid ethyl ester extracted from Hedyotic diffusa could induce apoptosis of leukemic cell line HL-60/ADR with muhidrug resistance, and the apoptotie rate was affected with dosage and time. Conclusion Hedyotic diffusa can effectively inhibit the proliferation of leukemic cell line HL-60/ADR with multidrug resistance, while inducing HL-60/ADR to apoptosis is one of its mechanism.
