摘要
①目的探讨转化生长因子-β(TGF-β)基因多态性与矽肺的关系。②方法采用1:1配对的病例对照研究方法,矽肺组为确诊的77例Ⅰ期矽肺患者,对照组与矽肺组同性别、同民族、同一工作车间或相同采掘工作面,年龄差<5岁,开始接尘时间和累积接尘工龄差<不超过2年的接尘工人。应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对TGF-β启动区的-509C/T位点和第一外显子的+869T/C、+915G/C位点的基因多态性进行检测。③结果矽肺组TGF-β启动区的-509C/T位点等位基因和基因型频率与对照组比较,差异均有显著性(P<0.05);+869T/C位点等位基因和基因型频率与对照组比较,差异均无显著性(P>0.05);+915G/C位点等位基因和基因型频率与对照组比较,差异均有显著性(P<0.05)。④结论TGF-β-509TT和+915GC基因型是矽肺的易感基因型;同时携带TGF-β-509*T和TGF-β+915*C等位基因的接尘人群,更易感矽肺。未发现TGF-β+869位点基因多态性与矽肺易感性有关联。
Objective To explore the association between gene polymorphisms of transforming growth factor-β(TGF -β) and susceptibility to silicosis. Methods The case - control study in 1 : 1 matching was used. The patients with silicosis (77 cses) were selected as case groups and the workers exposed to dust but not suffered from silicosis were regarded as the control group , who was the same sex, nationality, and workshop or work site as the case group and the differences of age and cumulative exposure time between the case and control group were not over five years and two years respectively. Polymerase chains reaction - restriction fragment length polymorphism ( PCR - RFLP) was used to research the frequencies of TGF -β genes in the two groups. Results The frequencies of the TGF -β(-509) genotypes and alleles in the case group were significantly different from in the control group ( P 〈 0.05 ). There was no significant difference in the frequencies of TGF -β + 869 genotypes and allelic between case group and control group ( P 〉 0.05 ) ; The frequencies of the TGF -β ( + 915 ) genotypes and aUeles in case group were significantly different from that in the control subjects( P 〈 0, 05). Conclusions TGF-β - 509TF genotype and + 915GC genotype may be a suscepti- ble factor to silicosis. The worker of carring TGF-β -509 * T and TGF -β + 915 * C alleles are more susceptible to silicosis. The T869C polymorphisms of the TGF-β gene are not associated with the silicosis ;
