摘要
①目的探讨胶原性关节炎大鼠滑膜组织中VEGF和NF-κB的mRNA的表达及其意义。②方法实验采用Ⅱ型胶原皮下多点注射的方法建立胶原诱导关节炎模型并进行关节指数评价。应用RT-PCR技术检测VEGF和NF-κB mRNA表达。③结果大部分大鼠在胶原致敏后2周左右发病,VEGF和NF-κB mRNA在模型组中的表达明显高于正常组,且两者的表达呈明显正相关。④结论VEGF和NF-κB参与了胶原性关节炎滑膜血管翳的形成过程,在RA的发病机制中具有重要意义。
Objective To investigate the mRNA expression and significance of vascular endothelial growth factor(VEGF) and nuclear factor of kappa B( NF -κB) in synovial tissues of rats with collagen induced arthritis(CIA). Methods The experimental arthritis model was established by subcutaneous injection of Ⅱ collagen and arthritis indexes were evaluated. The mRNA expressions of VEGF and NF -κB were detected with RT - PCR techniques. Results Most arthritis took place in 2 - 3 weeks after collagen injection. The expressions of VEGF and NF -κB in the model rats were higher than those in normal rats and there was a significant positive relation between VEGF and NF -κB. Conclusion The expression of VEGF and NF -κB is relative to the synovial pannus formation in CIA rats and may play an important role in RA.
