摘要 目的:研究高脂饮食对大鼠脂肪组织HSL和perilipin蛋白表达及脂肪水解的影响。方法:将116只3周龄SD雄性大鼠随机分为普食组(C组)和高脂组,10周后形成高脂饮食诱导肥胖大鼠(OB组)和肥胖抵抗大鼠(OR组)。测试大鼠形态、血清、脂肪组织HSL和perilipin蛋白表达等指标。结果:OB组大鼠体重、脂肪重量、血清FFA和甘油等明显高于C组和/或OR组;OB组大鼠脂肪组织HSL活性和蛋白表达显著低于C组和OR组;而OB组大鼠perilipin蛋白表达显著高于C组,但显著低于OR组。结论:高脂饮食导致大鼠肥胖与其脂肪组织HSL活性和蛋白表达下调以及perilipin蛋白表达上调,因而与促进脂肪储存有关。 Objective: Research the effect of high - fat diet on hormone sensitive lipase and perilipin protein expression as well as the lipolysis of rat adipose tissue. Methods : 116 SD male rats of three - week old are randomly divided into general diet control group (C Group) and high -fat diet group. After ten weeks, high -fat food is used to create obese rats (OB Group) and obesity- resistant rats (OR Group). The morphology, serum and adipose tissue HSL and perilipin are tested. Results: The weight, fat weight, serum FFA and serum glycerol are significantly higher than those in rats of C group or OR group; the adipose tissue HSL activity and protein expression of OB group rats are markedly lower than those in the C group; while the adipose tissue perilipin protein expression of OB group are markedly higher than those in C group, but significantly lower than those in OR group. Conclusion : The mechanism of obese rats induced by high - fat diet may be involved in the down - regulation of HSL activity and protein expression as well as the up - regulation of perilipin protein expression which promote fat storage.
