摘要
目的:研究异种无细胞真皮的组织相容性及血管化速度,并将结果与异体无细胞真皮的相关结果进行比较分析。方法:制备巴马香猪无细胞真皮和大鼠无细胞真皮,将二者同时皮下埋植于同一大鼠受体,1、4、8、12 w后取材,组织学切片观察其组织相容性;将二者分别进行单纯无细胞真皮移植,1、2、3、4 w后分别取材,组织学切片观察其血管化速度。结果:异种无细胞真皮与异体无细胞真皮皮下埋植后各时间点组织学反应相似,未引起受体免疫排斥反应,胶原结构未降解;单纯无细胞真皮移植后,异种无细胞真皮与异体无细胞真皮均能在2 w内完成血管化进程,且二者之间同一时间点新生血管数量无显著差异。结论:异种无细胞真皮与异体无细胞真皮同样具有组织相容性好、血管化速度快等生物学特性,理论上可替代异体无细胞真皮应用于临床。
Objective: To study of tissue compatibility and neovascularization of transplanted xeno - acellular dermal matrix ( ADM ) and compare the results with those of allo - ADM. Methods:The Bama miniature pig's ADM and Wistar rat's ADM were prepared and implanted in wounds of the same rat~ dorsum. After 1,4, 8, 12 w, biopsy specimens from each wound were stained with HE to evaluate tissue compatibility of xeno - ADM and allo - ADM. At the same time, xeno - ADM and allo - ADM were transplanted to the same rat's full thickness skin defects, and then new vessels in each biopsy specimen from the wounds were counted under microscope at 1, 2, 3, 4 w after implantation, respectively. Results: Histological reactions of xeno - ADM were similar to that of allo - ADM at all time points after implantation. Both materials did not cause immunological rejection and degradation of collagen structure. All of new vessels in xeno - ADM and allo - ADM were formed in 2 w after transplantation, and there was no significant difference in the quantity of new vessels between xeno - ADM and allo - ADM at the same time point. Conclusion : Xeno - ADM exhibits two important properties like allo - ADM: well tissue compatibility and rapid neovaseularization. It may be an ideal substitution of allo - ADM in clinical application.
