摘要 [目的]探索影响桂花愈伤组织诱导及增殖的激素组合。[方法]以紫银桂的幼叶、茎尖和幼胚为材料,对影响桂花愈伤组织形成和继代培养的激素组合进行研究。[结果]叶片、茎尖和幼胚在不同培养基中均能产生愈伤组织。在诱导培养基中NAA浓度为0.3 mg/L的条件下,当幼胚、幼叶和茎尖培养基中6-BA浓度分别为3.0、5.0和5.0 mg/L时,愈伤组织的诱导率最高。在诱导培养基中NAA浓度为0.5 mg/L的条件下,当幼胚、茎尖和幼叶培养基中6-BA浓度为3.0、3.05、.0 mg/L时,愈伤组织的诱导率最高。从节省成本的因素考虑,最适合愈伤组织增殖的激素组合应为6-BA 2.0+NAA 0.5 mg/L。[结论]该研究为桂花愈伤组织的诱导和增殖的进一步研究奠定了基础。 [Objective]The aim was to explore the hormone combination effecting the induction and proliferation of the callus from Osmanthus fragrans.[Method]With the young leaves,shoot-tips and immature embryos of O.fragrans as materials,the hormone combination effecting the formation and subculture of the callus from O.fragrans was studied.[Result] The young leaves,shoot-tips and immature embryos all could produce callus in different mediums.Under the condition that NAA was 0.3 mg/L in the induced medium,the induction rate of callus was highest when the 6-BA in the immature embryo,young leaf and shoot-tip mediums were 3.0,5.0 and 5.0 mg/L resp.Under the condition that NAA was 0.5 mg/L in the induced medium,the induction rate of callus was highest when the 6-BA in the immature embryo,shoot-tip and young leaf mediums were 3.0,3.0 and 5.0 mg/L resp.From the factor of saving cost,the optimum hormone combination for callus proliferation was 6-BA 2.0+NAA 0.5 mg/L.[Conclusion]The research laid the foundation for the further study of the induction and proliferation of the callus from O.fragrans.
