摘要
目的探讨微载体培养人肝细胞用于人工肝的可能性。方法用微载体 Cytodex 3培养人肝细胞,将其置于中空纤维型生物反应器构成人工肝装置,在体外循环实验中进行生物功能及细胞活力的观察与评价。结果体外两步法分离的人肝细胞,在辅助促聚条件下有效地粘附于微载体上,呈悬浮生长状。移人生物反应器内进行体外循环实验6小时,见循环液中白蛋白含量明显增加,利多卡因浓度显著减少,实验结束时肝细胞仍保持良好活力。结论微载体培养人肝细胞为材料的人工肝,具备生物合成与转化代谢功能,可望对肝衰竭患者起支持与治疗作用。
Objective To study the possibility of using microcarriers cultural human hepatocytes for artificial liver.Methods The biologic function of artificial liver device,consisting of microcarriers cultural human hepatocytes and a hollow fiber bioreactor was observed under circulating in vitro.Results On the conditions of facilitating aggreate,the human hepatocytes isolated by extracorporeal two-stage method were successfully attached to cytodex 3 microcarriers,which grew as suspending in media,setting the hepatocytes into the bioreactor and circulating the media for 6 hours,a markedly increase of ablumin and decrease of Lidocaine were found.A good viability of hepatocytes was also observed after the ex- periment.Conclusion The artificial liver with microcarriers cultural human hepatocytes has both the biosynthesis and biotransformation function.It could be expected to support and treat the patients of hepatic failure.
