摘要
目的研究莨菪亭(Scopoletin)祛暑功效及其机制。方法采用人工湿热环境热应激法复制家兔中暑动物模型,观察莨菪亭对热应激家兔平均发热曲线、到达42℃的时间均数及体温平均上升速度,研究其祛暑功效;应用比色法和滴定法测定家兔热应激1h后,心、脑、肾等组织中Na^+-K^+-ATPase的活性以及肺组织和血清中PLA2的活性,研究其祛暑机制。结果莨菪亭能显著延迟家兔在人工湿热环境中肛温到达42℃的时间(P<0.01~0.001);且明显降低单位时间肛温上升速度(P<0.001);于热应激前给予莨菪亭能有效地抑制心、脑、肾组织中的Na^+-K^+-ATPase活性,以及明显抑制血清、肺组织中的PLA2活性,这些作用与用药剂量呈明显的正相关(P<0.01)。结论莨菪亭通过抑制心、脑、肾组织中的Na^+-K^+-ATPase活性和血清。
Objective To study the effect of reducing summer-heat and its mechanisms of Scopoletin. Methods The temperature changes of heat-stressed or normal rabbits were observed after intragastric administrating Seopoletin. At the same time, the activity of Na^+ - K^+ - ATPase in heart, brain and kidney, the activity of PLA2 in serum and lung were measured by eolorimetry or titration. Results Scopoletin could delay the time of reaching 42 ℃ ( P 〈 0. 01 - 0. 001 ) and lower the rising rate of the core temperature (Trr) (P 〈 0. 001 )in heat-stressed rabbits in a dose-dependent manner respectively. It also could inhibit significantly the activity of Na ^+ - K ^+ - ATPase and PLA2 in the Heat-stressed rabbits( P 〈 0. 01 ). Conclusion : Inhibited the activity of Na^ + - K ^+ - ATPase in heart, brain, kidney and the activity of PLA2 in lung, serum were the mechanisms of reducing summer-heat effect of Seopoletin.
