巯基噻唑啉碱水解合成半胱胺盐酸盐

Synthesis of cysteamine hydrochloride via alkali hydrolyzation of mercaptothiazoline

摘要 研究了一种合成半胱胺盐酸盐的新方法,即由α-巯基噻唑啉碱水解合成半胱胺盐酸盐(CSH)的方法和条件。结果表明,常压碱水解能合成CSH,并确定了适宜条件为:氢氧化钠质量浓度30%,氢氧化钠与α-巯基噻唑啉的物质的量比为7∶1,水解压力0.3 MPa,水解时间3 h,CSH最高收率66.7%,纯度可达98.2%。合成的产物用核磁共振和红外光谱对结构进行标定。 In this paper, a new method and conditions for synthesis of cysteamine hydrochloride (CSH) by alkali hydrolyzation at atmospheric pressure were investigated. The results showed that the rate of alkali hydrolyzation at atmospheric pressure was much faster than the rate of hydrolyzation at high pressure, and the target product could be made by alkali hydrolyzation. The optimal conditions for alkali hydrolyzation were as follows:the mole ratio of 2-mereaptothiazoline to sodium hydroxide was 1:7;the mass concentration of sodium hydroxide was 30% ;and the distillation time was 3 h. The purity of CSH made by alkali hydrolysis was 98.1% , with the yield reaching 72.4% . The yield of CHS via al- kali hydrolysis of mercaptothiazoline at atmospheric pressure was 66.7%. The structure of the product was characterized by ^1HNMR and IR and the analytical results proved the product to be CSH.
