白花蛇舌草悬浮细胞摇瓶补料对多糖量的研究

摘要 通过摇瓶实验,研究补料方式对白花蛇舌草细胞悬浮培养细胞生长及多糖产量的影响。以细胞干重、多糖量为指标,对白花蛇舌草悬浮细胞进行了补糖(蔗糖)、补氮(氨水)和补碱(氢氧化钠)的摇瓶补料分批发酵实验。结果表明,4次补氮时白花蛇舌草悬浮细胞干重及多糖分别达到3.50 g/L和64.59 g/L;4次补碱时白花蛇舌草悬浮细胞干重及多糖产量分别达到3.27 g/L和58.87 g/L。补碱和补氮无论是对细胞干重还是多糖的产量等都有着明显的提高,说明它们对白花蛇舌草细胞悬浮培养细胞生长影响较大。 The prupose is to study on cell growth and polysaccharides of Hedyotis Diffusa Will's suspension cell culture by Flask shaking experiments. Flask-shaking fed batch fermentation is carried out by feeding sugar, nitrogen and alkali to it, according to cell dry weight (CDW) and the polysaccharides yield. By four feeding nitrogen, CDW and the polysaccharides yield reached 3.50 g/L,64.59 g/L respectively in Hedyotis Diffusa Will's suspension cell; by four feeding alkali,CDW and the polysaccharides yield reached 3.27 g/L, 58. 87 g/L in Hedyotis Diffusa Will's suspension cell respectively. CDW and the polysaccharides yield of Hedyotis Diddusa Will Cell are improved obviously by feeding nitrogen and alkali. It was obviously beneficial to Hedyotis DifJusa Will's suspension cell.
