表皮生长因子受体单克隆抗体对人肺腺癌细胞的生长抑制作用 被引量:2

The growth inhibition of anti EGF receptor monoclonal antibody to human lung adenocarcinoma cells

摘要 目的观察表皮生长因子受体(EGFR)单克隆抗体对人肺癌细胞体外增殖的作用,探讨其在肺癌抗体生物治疗中的应用价值。方法应用杂交瘤技术制备EGFR单克隆抗体并以噻唑蓝(MTT)比色法检测EGFR单克隆抗体对人肺癌细胞体外增殖能力的影响。结果EGFR单克隆抗体对人肺腺癌SPCA1和A549细胞体外增殖有显著的抑制作用,但对小细胞肺癌SH77和NCIH446细胞的生长无明显影响。抗体浓度为15μg/ml时,SPCA1和A549细胞的生长率仅为对照组的6%和12%。 Objective Epidermal growth factor (EGF) and its receptor play a critical role in the growth and regulation of many type of malignant cell. The high level of expression of the EGFR on nonmall cell lung cancer(NSCLC) and the important role of EGFR in signal transduction make it potentially an excellent target for antibody directed therapy.The purpose of this study was to evaluate the anti tumor effects of anti EGFR monoclonal antibody on lung cacinoma cells.Method Preparation of hybrid cell producing anti EGFR monoclonal antibody using EGF receptors partially purified from A431 cells as immunogen.The effect of anti EGFR monoclonal antibody to human lung cells was investigated in vitro.Result The anti EGFR monoclonal antibody was shown to inhibit the proliferation of human lung adenocarcinoma cells in vitro,but no growth inhibitory effect was observed in the human small cell lung cancer cells.The concentration,15 ug/ml,of anti EGFR antibody could inhibit growth rate by 94% and 88% for human lung adenocacinoma cell lines SPC A 1 and A549, respectively. Conclusion Our initial results indicate that the anti EGF receptor monoclonal antibody may be potentially useful for the biotherapy of lung adenocarcinoma.
