摘要
目的研究腺嘌呤用于制作雄性大鼠不育症模型的理想剂量和时效关系。方法将30只SD雄性大鼠分成3组(n=10),低剂量组每日予腺嘌呤300mg/kg灌胃给药,连续30d;高剂量组予腺嘌呤500mg/kg灌胃给药,连续15d;空白对照组予等量0.9%氯化钠溶液灌胃,连续30d。观察大鼠体征变化、睾丸系数、血清性激素水平及睾丸组织形态学改变。结果低剂量与高剂量腺嘌呤均能造成大鼠生殖功能损害,服用低剂量30 d或高剂量15d均造模成功。结论腺嘌呤对雄性大鼠生殖系统有明显毒性作用,应用高剂量可缩短造模时间。
Objective To explore the optimal dosage and duration in establishing adenine-induced male infertility rat model. Methods SD male rats were divided into 3 groups: in low dose groups rats were given adenine 300 mg/kg daily by intragastric administration for 30d, in high dose group were given 500 mg/kg daily for 15d, and in control group 0.9% saline was given. Observations were made on the appearance and activities, serum testosterone levels, weight index and histological changes of the testis. Results The sexual dysfunction was induced in male rats of low dose and high dose groups. Conclusion Adenine can induce the sexual dysfunction in male rats; and high dosage may reduce the induction duration.
